==== Type 2 ==== The SCFAs excreted by the soluble dietary fiber-consuming bacteria in the intestine activate FFAR2 on nearby intestinal L-cells. This stimules these cells to secrete GLP-1 (i.e., glucagon-like peptide-1) and PYY (i.e., peptide YY) into the blood. GLP-1 stimulates pancreatic beta cells to secrete insulin into the blood and inhibits pancreatic alpha cells from secreting glucagon into the blood. Since insulin causes cells to take up blood glucose and glucagon causes the liver to release glucose into the blood, FFAR2 activation of L cells lowers blood glucose levels. In addition, PYY and GLP-1 reduce appetite and food consumption. The excreted SCFAs also activate FFAR2 on nearby intestinal K cells to simulate their secretion of GIP (i.e., glucose-dependent insulinotropic polypeptide). GIP stimulates insulin secretion but, perhaps paradoxically, also stimulates glucagon secretion; however, the net effect of GIP is to reduce blood glucose levels. GIP also slows gastric motility. In addition, both GLP-1 and GIP protect pancreatic beta cells from dying by apoptosis (see programmed cell death). The SCFAs excreted by the gut microorganisms also pass through the intestinal epithelium to enter the blood stream and activate FFAR2 on cells located in distant tissues such as pancreas beta cells and adipose tissue fat cells. Individuals with type 2 diabetes, particularly in advanced cases, have nearly completely lost the incretin effect.
== Chemistry == Methylephedrine, also known as (1R,2S)-β-hydroxy-N,N-dimethyl-α-methyl-β-phenethylamine and as (1R,2S)-β-hydroxy-N,N-dimethylamphetamine, is a substituted phenethylamine and amphetamine derivative. It is the N-methylated derivative of ephedrine ((1R,2S)-β-hydroxy-N-methylamphetamine). A closely related compound and stereoisomer of methylephedrine is N-methylpseudoephedrine. Another related analogue is dimethylamphetamine (N,N-dimethylamphetamine), which is said to be a prodrug of methamphetamine and/or amphetamine. Other analogues include etafedrine (N-ethylephedrine) and cinnamedrine (N-cinnamylephedrine). The experimental log P of methylephedrine is 2.47 and its predicted log P ranges from 1.7 to 1.74. In organic chemistry, N-methylephedrine is used as a resolving agent and as a precursor to chiral supporting electrolytes, phase-transfer catalysts, and reducing agents.
== Side effects == Common side effects of leuprorelin injection include redness/burning/stinging/pain/bruising at the injection site, hot flashes (flushing), increased sweating, night sweats, tiredness, headache, upset stomach, nausea, diarrhea, impotence, testicular shrinkage, constipation, stomach pain, breast swelling or tenderness, acne, joint/muscle aches or pain, trouble sleeping (insomnia), reduced sexual interest, vaginal discomfort/dryness/itching/discharge, vaginal bleeding, swelling of the ankles/feet, increased urination at night, dizziness, breakthrough bleeding in a female child during the first two months of leuprorelin treatment, weakness, chills, clammy skin, skin redness, itching, or scaling, testicular pain, impotence, depression, or memory problems. The rates of gynecomastia with leuprorelin have been found to range from 3 to 16%. A cohort of women that were prescribed leuprorelin to delay precocious puberty as children has developed osteoporosis and brittle teeth at an unexpected rate; However, the FDA has not established that these conditions were caused by leuprorelin.
== Awards == Phillips has been honored by Paul Newman and by John F. Kennedy Jr. as one of America's most generous business leaders when EAS was honored in 1999 with a Newman's Own/George Award. == Treatments == Prophylaxis and treatment with an anti-inflammatory agent may stop progression of the reaction. Oral aspirin or ibuprofen every four hours for a day or 60 mg of prednisone orally or intravenously has been used as an adjunctive treatment . However, steroids are generally of no benefit. Patients must be closely monitored for the potential complications (collapse and shock) and may require IV fluids to maintain adequate blood pressure. If available, meptazinol, an opioid analgesic of the mixed agonist/antagonist type, should be administered to reduce the severity of the reaction. Anti TNF-α may also be effective.

===== Treatment ===== For people with cocaine abuse, mild symptoms may resolve completely with total abstinence from cocaine, making early involvement of addiction services essential. Repair may involve rhinoplasty, which includes creating a new internal lining with nasolabial flaps and restoring support with costal cartilage grafts. === Absorption === The bioavailability of meloxicam is decreased when administered orally compared to an equivalent IV bolus dose. Different oral formulations of meloxicam are not bioequivalent. Use of oral meloxicam following a high-fat breakfast increases the mean peak drug levels by about 22%; however, the manufacturer does not make any specific meal recommendations. In addition, the use of antacids does not show pharmacokinetic interactions. With chronic dosing, the time to maximum plasma concentration following oral administration is approximately 5–6 hours.
In 1795, observing the toll that poor nutrition took on his men, Napoleon announced a prize of 12,000 francs to anyone who could improve upon the prevailing food preservation methods of the time. In 1810, Nicholas Appert claimed the prize after devising a method of preserving food in which tightly sealing food inside a bottle and maintaining it at high temperature for certain period of time made the food safe for consumption until opening, using glass jars sealed with corks and boiling water. A trial in which food preserved by this method was sent overseas with French troops succeeded. Appert was given the money on condition that he make his discovery public, and duly published The Art of Preserving Animal and Vegetable Substances. The French Navy began using his methods. The Grande Armée experimented with issuing canned foods to its soldiers. However, the slow process of canning and the even slower development and transport stages prevented large amounts from being shipped across the French Empire, and the wars ended before the process was perfected. A major difficulty that presented itself was the use of glass, which was heavy, fragile, and could explode under internal pressure. Appert's work was improved upon by Philippe de Girard in 1811, when he pioneered a method of preserving food in metal cans. It was patented in Britain on his behalf by Peter Durand, who took the credit as its inventor. Girard, a Frenchman, preferred the entrepreneurial environment of Britain, but could not have taken out a patent in a country with which his own country was at war with.
==== Recent advances ==== Since the early 2000s, advances in droplet-based microfluidics have made it a powerful technique for conducting directed evolution campaigns. Early developments in bulk production of single-emulsions (SEs; e.g. "water-in-oil" droplets) and double-emulsions (DEs; e.g. "water-in-oil-in-water" droplets) were followed by innovations in on-chip formation and sorting of SEs and DEs, which allow for greater ease and throughput of directed evolution experiments on microfluidic chips. An essential component of directed evolution is the maintenance of the linkage between enzymatic genotypes and phenotypes. The ability to form DEs on-chip and subsequently sort using fluorescence-activated cell sorting (FACS) pushed the field forward. In 2013, Yan et al. showed the use of FACS to sort DEs. In 2014, Zinchenko et al. published a system to formulate monodisperse DEs and to sort and quantitatively analyze them using a commercially available flow cytometer. The authors demonstrated the power of their system by enriching an active wild-type arylsulfatase from populations of 0.1% and 0.01% active cells by 800- to 2500-fold, respectively. In 2016, Larsen et al. developed a fluorescence-based optical sorting system to monitor polymerases activity inside a microfluidic device. Using their system, Larsen and colleagues showed approximately 1200-fold enrichment of an engineered polymerase.
