This is a working overview of pentapeptide, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-02-01 and is reviewed periodically as new material appears.
Long-term storage of the dry powder is typically at minus twenty degrees Celsius or lower, protected from light and moisture. Solutions are commonly kept frozen and thawed only once, because repeated freeze-thaw cycles can promote aggregation and loss of measurable peptide content. Buffers near neutral pH are preferred over strongly acidic or strongly basic conditions. Shipping at ambient temperature is acceptable for short periods when the powder remains sealed and desiccated.
Identity and purity are assessed by complementary methods rather than a single test. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and reports a percentage purity. Mass spectrometry, most often with electrospray ionization, confirms the expected molecular mass and detects sequence-related variants. Amino acid analysis can verify composition, while water content and residual counterion measurements support the mass balance of a batch. Stability studies under accelerated conditions are used to estimate shelf life, though such estimates carry uncertainty for long-term storage.
One distinguishing feature reported in animal studies is selectivity. Ipamorelin stimulated growth hormone release with limited elevation of adrenocorticotropic hormone or cortisol compared with earlier secretagogues such as GHRP-6. This pattern has been described as more selective for the growth hormone axis. The finding comes mainly from preclinical work, and the degree to which it holds across species and doses is not fully settled. Reports also describe effects on gastric motility in animal models, suggesting activity outside the pituitary, though the clinical relevance of this observation is uncertain.
Ipamorelin is a synthetic pentapeptide that acts on the growth hormone secretagogue receptor, also known as the ghrelin receptor. Its sequence contains five amino acid residues, including a non-natural residue that increases stability against enzymatic breakdown. The compound was developed in the 1990s as part of research into small peptides that stimulate pituitary hormone release. Unlike larger protein hormones, it can be produced by solid-phase peptide synthesis and characterized by standard analytical methods.
At the receptor level, ipamorelin binds GHS-R1a and triggers signaling through Gq-coupled pathways. Activation leads to calcium release and downstream effects in pituitary somatotroph cells. These events promote the release of growth hormone into circulation. The response depends on the presence of the receptor and on the physiological state of the animal or tissue studied. Because the receptor is also found in other tissues, effects beyond the pituitary have been examined in laboratory models, though the extent of those effects remains an area of ongoing study.

Activity is mediated mainly through the growth hormone secretagogue receptor, now generally called the ghrelin receptor or GHS-R1a. Binding at this G-protein-coupled receptor triggers phospholipase C signaling, calcium mobilization, and release of growth hormone from pituitary somatotrophs. Reports describe less pronounced stimulation of adrenocorticotropic hormone and prolactin compared with earlier secretagogues such as hexarelin or GHRP-6. Selectivity figures vary between assay systems, so the degree of separation from other secretagogues is an area of ongoing comparison rather than a fixed constant.
In animal and early human studies, ipamorelin produces pulsatile growth hormone release and a secondary rise in insulin-like growth factor 1. The magnitude and duration of that rise depend on route, sampling schedule, and the baseline endocrine state of the subject. Whether repeated exposure alters the response over time is not firmly settled, since some reports describe stable pulsatility while others note attenuation. Most published data come from small samples, which limits the strength of any general claim about long-term behavior.
The Druze strictly avoid iconography, but use five colors ("Five Limits" خمس حدود khams ḥudūd) as a religious symbol: green, red, yellow, blue, and white. The First limit (green), identified by Ismail at-Tamimi (d. 1030) in the Epistle of the Candle, represents either Hamza Ibn Ali or Jesus, symbolizing the "Universal Mind/Intelligence/Nous".
The cells of fungi have in addition to the shared eukaryotic organelles a spitzenkörper in their endomembrane system, associated with hyphal tip growth. It is a phase-dark body that is composed of an aggregation of membrane-bound vesicles containing cell wall components, serving as a point of assemblage and release of such components intermediate between the Golgi and the cell membrane. The spitzenkörper is motile and generates new hyphal tip growth as it moves forward. The cell walls of fungi are uniquely made of a chitin-glucan complex. Chloroplasts are not found in fungal cells, their pigments are instead associated with their cell walls.
The two substrates of this enzyme are D-glyceric acid and oxidised nicotinamide adenine dinucleotide (NAD+). Its products are 2-hydroxy-3-oxopropanoate, reduced NADH, and a proton. The enzyme can alternatively use nicotinamide adenine dinucleotide phosphate as its cofactor. This enzyme belongs to the family of oxidoreductases, specifically those acting on the CH-OH group of donor with NAD+ or NADP+ as acceptor. The systematic name of this enzyme class is (R)-glycerate:NAD(P)+ oxidoreductase. This enzyme is also called tartronate semialdehyde reductase. This enzyme participates in glyoxylate and dicarboxylate metabolism.
== The Strep-tag principle == Just like other short-affinity tags (His-tag, FLAG-tag), the Strep-tag can be easily fused to recombinant proteins during subcloning of its cDNA or gene. For its expression, various vectors for various host organisms (E. coli, yeast, insect, and mammalian cells) are available. A particular benefit of the Strep-tag is its rather small size and the fact that it is biochemically almost inert. Therefore, protein folding or secretion is not influenced and usually it does not interfere with protein function. Strep-tag is especially suited for analysis of functional proteins, because the purification procedure can be kept under physiological conditions. This not only allows the isolation of sensitive proteins in a native state, but it is also possible to purify intact protein complexes, even if just one subunit carries the tag. In the first step of the Strep-tag purification cycle, the cell lysate containing Strep-tag fusion protein is applied to a column with immobilized Strep-Tactin (step 1). After the tagged protein has specifically bound to Strep-Tactin, a short washing step with a physiological buffer (e.g. phosphate buffered saline, PBS) removes all other host proteins (step 2). This is due to Strep-Tactin's low tendency to bind proteins non specifically. Then, the purified Strep-tag fusion protein is gently eluted with a low concentration of desthiobiotin, which specifically competes for the biotin binding pocket (step 3). To regenerate the column, desthiobiotin is removed by application of a HABA containing solution (a yellow azo dye).
In many-body potentials, the potential energy includes the effects of three or more particles interacting with each other. In simulations with pairwise potentials, global interactions in the system also exist, but they occur only through pairwise terms. In many-body potentials, the potential energy cannot be found by a sum over pairs of atoms, as these interactions are calculated explicitly as a combination of higher-order terms. In the statistical view, the dependency between the variables cannot in general be expressed using only pairwise products of the degrees of freedom. For example, the Tersoff potential, which was originally used to simulate carbon, silicon, and germanium, and has since been used for a wide range of other materials, involves a sum over groups of three atoms, with the angles between the atoms being an important factor in the potential. Other examples are the embedded-atom method (EAM), the EDIP, and the Tight-Binding Second Moment Approximation (TBSMA) potentials, where the electron density of states in the region of an atom is calculated from a sum of contributions from surrounding atoms, and the potential energy contribution is then a function of this sum.

== Synthesis == Phosphorimidazolide reagents have been synthesized from phosphate mono-esters. In one method, a phosphate mono-ester is dissolved in anhydrous pyridine or N,N-dimethylformamide (DMF) and activated using triphenylphosphine (PPh3) and 2,2’-Dithiodipyridine (2,2’-DTDP) in the presence of triethylamine (TEA) base and excess imidazole. In another method using fewer reagents, a phosphate mono-ester is dissolved in DMF and carbonyldiimidazole (CDI) is used to both remove an oxygen atom from the phosphate group and supply the imidazole substituent. The product of either reaction may be collected by precipitation using acetonitrile or acetone as antisolvent with sodium or lithium perchlorate to supply the sodium or lithium salt of the phosphorimidazolide respectively. Alternatively, the phosphorimidazolide may be isolated by reverse-phase flash column chromatography with TEAB buffer and acetonitrile.
=== Popular culture === Lithium has now become a part of Western popular culture. Characters in Maude, Pi, Homeland, Premonition, Stardust Memories, American Psycho, Garden State, and An Unmarried Woman all take lithium. Lithium is the chief constituent of the calming drug in Ira Levin's dystopian novel This Perfect Day. SiriusXM Satellite Radio in North America has a 1990s alternative rock station called Lithium, and several songs refer to the use of lithium as a mood stabilizer. These include: "Equilibrium met Lithium" by South African artist Koos Kombuis, "Lithium" by Evanescence, "Lithium" by Nirvana, "Lithium and a Lover" by Sirenia, "Lithium Sunset", from the album Mercury Falling by Sting, and "Lithium" by Thin White Rope.
In February 2019, Sarepta acquired five gene therapy candidates for $165 million after one of them, MYO-101, produced results with a new gene therapy candidate for patients with Limb-Girdle muscular dystrophy; two months after receiving a single treatment, muscles from all three patients were producing the protein they couldn't make on their own. In 2019, the FDA rejected a Serepta Duchenne muscular dystrophy drug before reversing course 3 months later. The company opened a new Genetic Therapies Center of Excellence in Columbus, Ohio in October, 2021 in order to dramatically advance research into therapeutics for several muscular dystrophies that had begun at Nationwide Children's Hospital several years earlier. As of 2022, there are three FDA-approved DMD drugs in Sarepta Therapeutics' portfolio. In January 2023, Sarepta partnered with Catalent to manufacture delandistrogene moxeparvovec (SRP-9001). In June 2023, the FDA approved Sarepta Therapeutics’ fourth therapy, delandistrogene moxeparvovec-rokl, the first gene therapy to treat Duchenne muscular dystrophy (DMD). The product was approved under the accelerated approval pathway for 4-5 year olds with a confirmed DMD diagnosis. Accelerated approval requires that the product be studied further to verify its clinical benefit. In July 2025, Sarepta announced plans to lay off 500 employees, about 36% of its workforce, following the deaths of two patients linked to its Duchenne muscular dystrophy gene therapy, ELEVIDYS.
Mutations that prevent the formation of MinE rings result in the MinCD zone extending well beyond the polar zones, preventing FtsZ to polymerize and to perform cell division. MinD requires a nucleotide exchange step to re-bind to ATP so that it can re-associate with the membrane after MinE release. The time lapse results in a periodicity of Min association that may yield clues to a temporal signal linked to a spatial signal. In vivo observations show that the oscillation of Min proteins between cell poles occurs approximately every 50 seconds. Oscillation of Min proteins, however, is not necessary for all bacterial cell division systems. Bacillus subtilis has been shown to have static concentrations of MinC and MinD at the cell poles. This system still links cell size to the ability to form a septum via FtsZ and divide.

=== The Deep Hot Biosphere === At the beginning of his 1992 paper The Deep Hot Biosphere, Thomas Gold referred to ocean vents in support of his theory that the lower levels of the earth are rich in living biological material that finds its way to the surface. He further expanded his ideas in the book The Deep Hot Biosphere. An article on abiogenic hydrocarbon production in the February 2008 issue of Science journal used data from experiments at the Lost City hydrothermal field to report how the abiotic synthesis of low molecular mass hydrocarbons from mantle derived carbon dioxide may occur in the presence of ultramafic rocks, water, and moderate amounts of heat.

It acts on the growth hormone secretagogue receptor, GHS-R1a, which is also the receptor for ghrelin. Binding triggers intracellular signaling that promotes growth hormone release from the pituitary. The interaction is the basis for its classification as a secretagogue.
